Answer PaperPaper Code: 245ABMaximum Marks: 75Time: 3 Hours

Note: i) Question paper consists of Part A, Part B. ii) Part A is compulsory, which carries 25 marks. In Part A, answer all questions. iii) In Part B, Answer any one question from each unit. Each question carries 10 marks and may have a, b as sub questions.

Part A

1a

Give the significance of amorphous forms in preformulation studies.

2 marks

Answer

Amorphous forms are significant in preformulation because they lack a long-range crystalline lattice structure, which results in higher thermodynamic energy and increased molecular mobility compared to crystalline forms. This leads to significantly enhanced apparent solubility and faster dissolution rates, which are critical for improving the bioavailability of poorly water-soluble drugs. However, their physical instability and tendency to revert to a more stable crystalline state require careful evaluation during the preformulation stage to ensure long-term shelf-life and performance.

1b

What is the influence of pKa and pH during preformulation studies?

3 marks

Answer

The pKa (dissociation constant) and pH of the environment are critical in preformulation because they dictate the ionization state of a drug molecule, which directly influences its solubility, stability, and permeability.

1. Solubility: According to the Henderson-Hasselbalch equation, the solubility of ionizable drugs is highly pH-dependent. Weak acids are more soluble in alkaline environments, while weak bases are more soluble in acidic environments due to the formation of soluble ionized salts.

2. Permeability: Only the unionized form of a drug can effectively partition through lipid biological membranes. The ratio of ionized to unionized drug, determined by the pKa and the pH of the physiological site (e.g., stomach vs. intestine), dictates the extent of passive absorption.

3. Stability: Many drugs undergo pH-catalyzed degradation (e.g., hydrolysis). Identifying the pH of maximum stability is essential for formulating stable liquid dosage forms and selecting appropriate buffering agents.
