PART - A

1a

Define antimetabolites. Give the structure of Azathioprine.

2 marks

Answer

Antimetabolites are structural analogs of normal metabolites that interfere with the synthesis of essential cellular components, such as DNA or RNA, by competing with natural substrates for enzymes or by being incorporated into nucleic acids, thereby inhibiting cell growth and division. They are primarily used as antineoplastic or immunosuppressive agents.

The structure of Azathioprine is 6-[(1-methyl-4-nitro-1H-imidazol-5-yl)sulfanyl]-9H-purine.

1b

Write a note on synthesis of Promethazine hydrochloride.

3 marks

Answer

Promethazine hydrochloride is synthesized starting from phenothiazine. The process involves the following steps:

  1. Phenothiazine is reacted with 2-chloropropyl dimethylamine in the presence of a strong base like sodium amide (NaNH2) in a non-polar solvent such as xylene or toluene.
  2. This reaction involves the N-alkylation of the phenothiazine nitrogen atom with the side chain.
  3. The resulting base, promethazine, is then treated with hydrochloric acid in an organic solvent like ether or ethanol to form the stable, crystalline promethazine hydrochloride salt.
Phenothiazine + Cl-CH2-CH(CH3)-N(CH3)2 → Promethazine base → Promethazine hydrochloride
