Explain about the nutritional requirements and raw materials used for culture media.
Answer
Culture media are essential for the growth, identification, and maintenance of microorganisms. To support microbial life, the medium must provide specific nutrients that mirror the organism's natural environment.
The fundamental nutritional requirements include:
- Water: Acts as the solvent for all chemical reactions and nutrient transport.
- Carbon Source: Essential for building cellular components; autotrophs use CO2, while heterotrophs require organic compounds like glucose or starch.
- Nitrogen Source: Required for the synthesis of proteins, nucleic acids, and enzymes. Sources include peptones, amino acids, or inorganic nitrates/ammonium salts.
- Energy Source: Usually provided by carbon compounds or light in photosynthetic organisms.
- Minerals and Trace Elements: Ions such as Mg2+, Fe2+, K+, and Ca2+ act as enzyme cofactors and maintain osmotic pressure.
- Growth Factors: Vitamins, purines, and pyrimidines that the organism cannot synthesize itself.
Common raw materials used in media preparation include:
- Peptone: A complex mixture of partially digested proteins providing nitrogen and amino acids.
- Meat Extract: Provides vitamins, minerals, and organic growth factors.
- Yeast Extract: A rich source of B-complex vitamins and amino acids.
- Agar: A polysaccharide derived from seaweed used as a solidifying agent; it remains liquid at high temperatures and solidifies at approximately 42°C.
- Blood/Serum: Used for fastidious organisms requiring enriched media.
Describe the phase contrast microscope.
Answer
A phase contrast microscope is a specialized optical instrument used to observe living, unstained microorganisms by converting phase shifts in light passing through a transparent specimen into brightness changes in the image.
Key components and working principle:
- Annular Diaphragm: Located below the condenser, it produces a hollow cone of light that illuminates the specimen.
- Phase Plate: Situated in the objective lens, this plate contains a phase ring that advances or retards the phase of the undeviated light (direct light) by 1/4 wavelength.
- Interference: Light passing through the specimen (diffracted light) and light passing around the specimen (direct light) recombine at the image plane. The phase difference between these two beams results in constructive or destructive interference, creating contrast.
This technique is superior for viewing live cells, motility, and internal structures like endospores or inclusion bodies without the need for chemical fixation or staining, which often kill or distort the specimen.
