List out the Advantages of Electron Microscope over Optical Microscope.

7 marks

Answer

The primary advantages of an electron microscope over an optical microscope stem from the use of electrons, which have a significantly shorter de Broglie wavelength than visible light, allowing for superior imaging capabilities:

  • Higher Resolution: Electron microscopes achieve sub-nanometer resolution, whereas optical microscopes are limited by the diffraction of light to approximately 200 nm.
  • Higher Magnification: Electron microscopes can provide magnifications up to 1,000,000x or more, compared to the typical limit of 1,000x to 2,000x for optical systems.
  • Depth of Field: Electron microscopes offer a much greater depth of field, allowing for the observation of three-dimensional structures with better focus across uneven surfaces.
  • Versatile Contrast Mechanisms: Beyond simple absorption, electron microscopes utilize secondary electrons, backscattered electrons, and characteristic X-rays to provide detailed information about topography, composition, and crystallography.
  • Analytical Capabilities: Integrated systems like EDS (Energy Dispersive Spectroscopy) allow for precise elemental analysis of the specimen, a feature not natively available in standard optical microscopy.

Construct a note on Resolution and Depth of Field of Optical Microscope.

Answer

Resolution in an optical microscope is defined as the minimum distance between two points at which they can still be distinguished as separate entities. It is governed by the Abbe diffraction limit, expressed by the formula:

d = 0.61λ / (n sin θ) = 0.61λ / NA

Where d is the resolution limit, λ is the wavelength of the light used, n is the refractive index of the medium, and θ is the half-angle of the cone of light entering the objective lens. The term n sin θ is known as the Numerical Aperture (NA). Improving resolution requires either decreasing the wavelength or increasing the numerical aperture.

Depth of Field refers to the axial distance through which an object can be moved while remaining in acceptable focus. It is inversely proportional to the square of the numerical aperture and the magnification of the objective lens. While a high NA is desirable for high resolution, it inherently results in a very shallow depth of field, meaning only a thin section of the specimen is in focus at any given time. Consequently, optical microscopy often requires thin sectioning of samples to obtain clear images, as thicker specimens will appear blurred outside the narrow focal plane.
