Explain the principle of the phase-contrast microscope with a neat diagram. Discuss the advantages and disadvantages of phase contrast microscope?

15 marks

Answer

The Phase-Contrast Microscope is an optical imaging technique that converts phase shifts in light passing through a transparent specimen into brightness changes in the image. Transparent specimens, such as living cells, often have very little contrast because they do not absorb light significantly. However, they do cause small phase shifts in the light wave due to differences in refractive index and thickness.

The principle relies on the interference of light waves. When light passes through a specimen, the light rays are either deviated (scattered) or undeviated. The microscope uses a phase plate located in the objective lens to advance or retard the phase of the undeviated light by a quarter-wavelength (λ/4). This creates a phase difference between the deviated and undeviated light, which then interfere constructively or destructively at the image plane, resulting in a visible contrast between the specimen and the background.

Diagram description: The light source passes through an annular diaphragm, creating a hollow cone of light. This light passes through the specimen. The undeviated light passes through the phase plate (a ring-shaped phase-shifting material), while the deviated light passes through the rest of the objective lens. Upon recombination at the image plane, the phase differences are translated into amplitude (intensity) differences.

Advantages:

  • Allows for the observation of living, unstained cells and tissues in their natural state.
  • Eliminates the need for chemical fixation or staining, which can kill or distort the specimen.
  • Provides high-contrast images of transparent objects that would be invisible under a standard bright-field microscope.

Disadvantages:

  • Halo effect: A bright ring often appears around the edges of the specimen, which can obscure fine detail.
  • Shading-off effect: Large, thick specimens may appear uniformly bright or dark, losing internal detail.
  • The equipment is more expensive and complex to align compared to basic bright-field microscopes.
  • Not suitable for very thick specimens as they cause excessive light scattering.

Differentiate between SEM and TEM in terms of working principle, sample preparation and applications.

15 marks

Answer

Scanning Electron Microscopy (SEM) and Transmission Electron Microscopy (TEM) are both powerful electron-based characterization techniques, but they differ significantly in their operational modes and the information they provide.

FeatureScanning Electron Microscopy (SEM)Transmission Electron Microscopy (TEM)
Working PrincipleA focused electron beam scans the surface of the specimen in a raster pattern. Secondary or backscattered electrons are detected to form an image of the surface topography.A broad electron beam passes through a very thin specimen. The transmitted electrons are focused by electromagnetic lenses to form an image based on diffraction and mass-thickness contrast.
Sample PreparationSamples must be conductive or coated with a thin layer of conductive material (e.g., gold/carbon). The sample can be relatively thick and bulkier.Samples must be extremely thin (typically less than 100 nm) to allow electron transmission. This requires complex preparation like electropolishing, ion milling, or ultramicrotomy.
ResolutionProvides high-resolution images of surface morphology and topography, typically in the range of 1-10 nm.Provides atomic-scale resolution, allowing for the visualization of crystal lattices, defects, and internal structure, often below 0.1 nm.
ApplicationsUsed for studying surface fracture surfaces, grain structure, surface coatings, and particle size distribution.Used for studying internal crystal structure, phase identification, dislocation density, and nanostructure analysis.

In summary, SEM is primarily a surface-sensitive technique used for morphological analysis, whereas TEM is a transmission-based technique used for detailed internal structural and crystallographic analysis at the atomic level.
