What is prodrug and write approaches to improve solubility and permeability with examples?
Answer
A prodrug is a pharmacologically inactive derivative of a drug molecule that undergoes biotransformation in vivo to release the active parent drug, typically to overcome limitations such as poor solubility, low permeability, or chemical instability.
Approaches to improve solubility and permeability include:
- Improving Solubility: This is achieved by attaching polar or ionizable groups to the parent molecule. For example, converting hydroxyl-containing drugs into phosphate or hemisuccinate esters increases water solubility. A classic example is the conversion of chloramphenicol to chloramphenicol sodium succinate for intravenous administration.
- Improving Permeability: This involves masking polar or ionizable groups to increase lipophilicity, allowing better passive diffusion across biological membranes. For example, converting carboxylic acid groups into simple alkyl esters or acyloxymethyl esters. An example is the conversion of the polar antibiotic enalapril into the more lipophilic prodrug enalaprilat, which is then hydrolyzed by esterases in the liver to the active form.
Write note on combinatorial chemistry.
Answer
Combinatorial chemistry is a high-throughput technology used to synthesize a large number of chemical compounds in a single process, creating a chemical library for rapid drug discovery. It enables the simultaneous preparation of thousands of diverse molecules by systematically varying building blocks.
Key aspects include:
- Solid-Phase Synthesis: Often utilizes resin beads as a support, allowing for easy removal of excess reagents and by-products through simple washing steps.
- Split-and-Mix Method: A core technique where a resin is divided into multiple reaction vessels, reacted with different building blocks, pooled together, and then re-divided for the next step, resulting in exponential growth of library diversity.
- Parallel Synthesis: Compounds are synthesized in individual, separate reaction vessels, which is useful for generating smaller, focused libraries.
This approach significantly accelerates the lead optimization phase in medicinal chemistry by allowing researchers to screen vast chemical spaces against biological targets efficiently.
