1

Write environmental conditions required for animal housing area as per CPCSEA guidelines.

15 marks

Answer

According to the CPCSEA (Committee for the Purpose of Control and Supervision of Experiments on Animals) guidelines, the animal housing facility must be designed and maintained to ensure the welfare of laboratory animals and the reliability of experimental results. The key environmental conditions are as follows:

  • Temperature and Humidity: The animal room temperature should be maintained between 18°C and 29°C. Relative humidity should be controlled within the range of 30% to 70%.
  • Ventilation: Adequate ventilation is mandatory to ensure a continuous supply of fresh air and to remove heat, moisture, and odors. A minimum of 10 to 15 air changes per hour is recommended for most laboratory animal species.
  • Lighting: A controlled lighting system is required, typically following a 12-hour light and 12-hour dark cycle. Light intensity should be sufficient for animal care and observation without causing stress or ocular damage to the animals.
  • Noise Control: The facility should be designed to minimize noise. Loud noises and vibrations must be avoided, as they can induce physiological stress, alter endocrine function, and affect the reproducibility of experimental data.
  • Sanitation and Hygiene: Regular cleaning and disinfection schedules must be strictly followed. Waste disposal must be managed to prevent contamination and the spread of pathogens.
  • Space Requirements: Animals must be housed in cages or pens that provide sufficient space for normal postural adjustments, grooming, and social interaction, adhering to the specific floor area and height requirements defined by CPCSEA for each species.
  • Environmental Enrichment: Cages should include appropriate enrichment materials (such as nesting material, hiding spots, or chew toys) to promote species-specific behaviors and improve animal well-being.
  • Monitoring: Continuous monitoring and documentation of these environmental parameters are essential to ensure compliance with ethical standards and to maintain the health status of the animal colony.
2

Explain limitations of animal experimentation and alternate animal experiments.

15 marks

Answer

Animal experimentation, while foundational to pharmacology, faces significant constraints that have driven the development of alternative methods. The limitations of using animal models include species-specific differences in pharmacokinetics and pharmacodynamics, which often lead to poor translation of results to human clinical outcomes. Furthermore, animals cannot report subjective experiences like pain, nausea, or psychological distress, complicating the assessment of certain drugs. Ethical concerns regarding animal welfare, the high cost of maintaining animal facilities, and the long duration required for chronic studies also limit their utility.

To address these challenges, the 3Rs principle (Replacement, Reduction, and Refinement) is employed, focusing on the following alternative methods:

  • In vitro models: Use of isolated cells, tissues, or organ cultures (e.g., cell lines, human primary cells, or organoids) to study drug mechanisms, toxicity, and metabolism without the need for live animals.
  • In silico methods: Computer-based modeling and simulation, such as Quantitative Structure-Activity Relationship (QSAR) analysis and molecular docking, which predict drug interaction, toxicity, and efficacy based on chemical structure and existing databases.
  • Organ-on-a-chip technology: Microfluidic devices that mimic the physiological environment and functional units of human organs (e.g., liver-on-a-chip or lung-on-a-chip), providing a more accurate representation of human systemic responses.
  • Human-based studies: Utilization of micro-dosing in human volunteers, where sub-therapeutic doses are administered to study pharmacokinetics using highly sensitive analytical techniques like Accelerator Mass Spectrometry (AMS).
  • Mathematical and statistical modeling: Advanced algorithms used to predict drug behavior and optimize experimental designs, thereby reducing the number of animals required for statistical significance.

These alternatives provide a more human-relevant, ethical, and often more cost-effective approach to drug discovery and safety assessment.
