Part A

1a

Write a short note about personnel requirement in pilot studies in pharmaceutical research.

2 marks

Answer

Personnel involved in pilot studies must possess a combination of specialized technical skills and a thorough understanding of both laboratory-scale and commercial-scale manufacturing processes. The team typically includes experienced pharmaceutical scientists, process engineers, and quality assurance personnel who collaborate to bridge the gap between R&D and full-scale production. These individuals are responsible for evaluating equipment suitability, optimizing process parameters, and ensuring that the pilot-scale operations strictly adhere to Current Good Manufacturing Practices (cGMP) to maintain product quality and safety.

1b

Define SUPAC and elaborate the guidelines for SUPAC.

3 marks

Answer

SUPAC stands for Scale-Up and Post-Approval Changes. It refers to guidelines provided by the FDA for pharmaceutical manufacturers to make changes in the formulation, manufacturing process, equipment, or site of production after the initial approval of a New Drug Application (NDA) or Abbreviated New Drug Application (ANDA).

The guidelines are designed to ensure that the quality, safety, and efficacy of the drug product remain unchanged despite these modifications. Key aspects of the guidelines include:

  • Classification of Changes: Changes are categorized into Level 1 (minor changes with minimal impact), Level 2 (changes with moderate impact), and Level 3 (major changes with significant impact).
  • Documentation Requirements: Depending on the level of change, manufacturers must provide specific documentation, such as stability data, dissolution testing, or bioequivalence studies, to support the change.
  • Regulatory Filing: The guidelines specify the type of regulatory submission required, such as an Annual Report, a Changes Being Effected (CBE) supplement, or a Prior Approval Supplement (PAS), based on the risk level associated with the modification.
