PART - A

1a

Give the structure and uses of Diphenhydramine HCL.

2 marks

Answer

The structure of Diphenhydramine hydrochloride is 2-(diphenylmethoxy)-N,N-dimethylethanamine hydrochloride. It consists of a benzhydryl ether group attached to an N,N-dimethylethylamine chain, forming a salt with hydrochloric acid.

C₁₇H₂₁NO · HCl

Its primary uses include the treatment of allergic conditions such as rhinitis, urticaria, and pruritus. It is also used as an antiemetic for motion sickness, a mild sedative, and as an antitussive agent.

1b

Give the mechanism of action of triprolidine hydrochloride.

3 marks

Answer

Triprolidine hydrochloride acts as a potent, competitive, and reversible H1-receptor antagonist. It functions by binding to the H1-histamine receptors on effector cells, thereby preventing the binding of endogenous histamine to these receptors.

By blocking the H1 receptors, triprolidine inhibits the physiological effects typically mediated by histamine, such as capillary permeability, smooth muscle contraction in the bronchi and gastrointestinal tract, and the stimulation of sensory nerve endings that lead to itching and sneezing. Because it is an inverse agonist, it also suppresses the constitutive activity of the H1 receptor, effectively reducing allergic symptoms associated with conditions like rhinitis and urticaria.
