a

Explain the Controlled Release of drugs from Ion – Exchange Resins.

8 marks

Answer

Ion-exchange resins are cross-linked, water-insoluble polymers containing ionizable functional groups that can exchange ions with surrounding medium. The controlled release of drugs from these resins is based on the principle of ion exchange, where a drug molecule is bound to the resin matrix via ionic interactions.

The process involves the following steps:

  1. Drug loading: The drug (either cationic or anionic) is loaded onto the resin by passing a drug solution through a column of resin or by batch equilibration, forming a drug-resin complex known as a resinate.
  2. Release mechanism: When the resinate is ingested, it encounters the gastrointestinal (GI) fluids containing high concentrations of electrolytes (e.g., H+, Na+, Cl-).
  3. Ion exchange: The ions present in the GI fluid exchange with the drug molecules bound to the resin matrix. The drug is displaced from the resin and released into the dissolution medium.
  4. Diffusion: The rate of release is governed by the diffusion of the drug out of the resin matrix and the diffusion of counter-ions into the matrix.

The release rate is primarily controlled by the degree of cross-linking of the resin, the particle size, and the concentration of electrolytes in the GI tract. By coating the drug-resin complex with rate-controlling polymers, the release can be further sustained to achieve a desired zero-order or first-order release profile.

b

Write about the process of polymeric degradation and Erosion.

7 marks

Answer

Polymeric degradation and erosion are critical processes in the design of biodegradable drug delivery systems, determining the rate and duration of drug release.

Polymeric Degradation: This refers to the chemical breakdown of polymer chains into smaller fragments, typically through the cleavage of covalent bonds in the polymer backbone. The primary mechanism is hydrolysis, where water molecules react with hydrolytically unstable linkages like esters, anhydrides, or amides. Degradation can be classified as:

  • Bulk erosion: Water penetrates the entire polymer matrix faster than the rate of degradation, causing the polymer to degrade throughout the bulk. This often leads to a sudden loss of structural integrity and a burst release of the drug.
  • Surface erosion: The rate of degradation at the surface is faster than the rate of water penetration into the bulk. The polymer thins from the outside in, which allows for more predictable, zero-order release kinetics.

Erosion: This is the physical process of mass loss from the polymer matrix. It is the consequence of degradation. As the polymer chains break down into soluble oligomers or monomers, they dissolve and diffuse out of the matrix, leading to physical erosion. The combined process of degradation and erosion dictates the drug release profile, as the drug is released either by diffusion through the degrading matrix or as a result of the matrix disappearing entirely.
