JNTU B.Pharmacy II PHARMACEUTICAL MICROBIOLOGY Previous Question Paper (October/November-2020 - R17)
Answer any five questions. All questions carry equal marks.
Main
Explain the scope and importance of microbiology.
Answer
Microbiology is the study of microorganisms, including bacteria, viruses, fungi, protozoa, and algae. Its scope and importance in pharmacy and medicine are extensive:
- Pharmaceutical Industry: Essential for the production of antibiotics, vaccines, vitamins, and enzymes through fermentation processes.
- Quality Control: Ensures the sterility and microbial safety of pharmaceutical products, preventing contamination that could lead to product degradation or patient harm.
- Clinical Diagnosis: Enables the identification of pathogenic microorganisms, facilitating the selection of appropriate antimicrobial therapy.
- Public Health: Contributes to the understanding of disease transmission, epidemiology, and the development of sanitation and sterilization protocols.
- Biotechnology: Provides the foundation for genetic engineering, allowing for the synthesis of complex therapeutic proteins like insulin.
- Environmental Microbiology: Studies the role of microbes in waste treatment and the degradation of environmental pollutants.
Write a note on the various preservation methods for pure cultures.
Answer
Preservation of pure cultures is vital to maintain the genetic stability and viability of microbial strains over long periods. Common methods include:
- Subculturing (Serial Transfer): Periodic transfer of cultures to fresh media. While simple, it risks genetic mutation and contamination over time.
- Refrigeration: Storing cultures at 0-4°C slows metabolic activity, extending the shelf life of cultures for several weeks or months.
- Paraffin Method: Covering an agar slant with sterile mineral oil prevents dehydration and restricts oxygen supply, slowing down metabolism.
- Cryopreservation: Storing cultures at extremely low temperatures (e.g., -80°C or in liquid nitrogen at -196°C) using cryoprotectants like glycerol or DMSO to prevent ice crystal formation, which is the most effective long-term method.
- Lyophilization (Freeze-drying): The removal of water from a frozen culture under vacuum. The resulting dry powder can be stored for years and is easily reconstituted, making it ideal for the long-term storage of industrial and clinical strains.
